OBJECTIVES The mechanism is not very clear during tumorigenesis of transitional cell carcinoma of urinary bladder. Recent investigation suggests a correlation among cyclooxygenase-2, tumorivasc- ularization and tumorigenesis. This study is designed to investigate the role of cyclooxygenase-2 and microvascular density in the tumorigenesis and progression of transitional cell carcinoma of urinary bladder,also evaluate their clinical significance in transitional cell carcinoma of urinary bladder.So as to look for new tumor markers and targets of treatment on transitional cell carcinoma of urinary bladder.METHODS With anti-cyclooxygenase-2 McAb and anti-VWF:Ag McAb in this study,immunohistochemistry was used to study cyclooxygenase-2 and the tumor interstitial vascular density in 56 cases of transitional cell carcinoma of urinary bladder.And as a contrast, 8 cases of normal bladder tissue and 12 cases of chronic cystitis tissue was collected. RESULTS The expression of cyclooxygenase-2 intransitional cell carcinoma of urinary bladder was positivelycorrelated to the tumor interstitial vascular density ;Both of them were related to the tumor pathologic grade,the invasiveness of the tumor tissue, the lymphatic transferation and recurrence time of tumor (P< 0.05) ,but not related to the tumor size or tumor number ( P> 0.05 ) .CONCLUSIONS 1. cyclooxygenase-2 wasoverexpressed in transitional cell carcinoma of urinary bladder and played an important role in oncogenesis,development and recurrence of transitional cell carcinoma of urinary bladder. 2.The expression of cyclooxygenase-2 was positively related to the tumorivascularization. 3.The overexpression ofcyclooxygenase-2 and high microvascular density had remarkable significance in evaluating the prognosis of transitional cell carcinoma of urinary bladder and the above two maybe a new target of medical treatment for transitional cell carcinoma of urinary bladder.
