Objective: The expression of P-gp and CX43 were studied in primary epithelial ovarian carcinoma(PEOC) and related differences between drug-resistance group and drug-sensitive group were analyzed and their clinical value in ovarian cancer drug-resistance, chemotherapic drug choice and patient's prognosis forecast was further investigated. Methods: 1.60 cases PEOC of Gynecologic Oncology Department of Jiangsu Cancer Hospital between January 1999 and June 2003 were studied retrospectively for their clinic data and pathologic data. Among 60 cases, 30 cases of chemotherapic drug-resistance and the rest 30 cases of chemotherapic drug- sensitive with additional 6 normal ovarian specimen as control.2.Pathological paraffin slices of 60 cases PEOC and 6 cases normal ovarian specimen were deparafFmized by xylol and were made the single cell suspension. P-gp and CX43 were quantitatively analysed by FCM. Results: 1. 30 cases of chemotherapic drug-resistance group: The expression of CX43 and P-gp were respectively 2.156±0.124 and 1.563±0.139. 2. 30 cases of chemotherapic drug-sensitive group: The expressions of CX43 and P-gp were respectively 2.237±0.188 and1.131±0.125o 3. 6 cases normal ovary group: The expression of CX43 and P-gp were respectively 2.255 + 0.189 and 0.823±0.189o 4. The study revealed that the CX43 expression was depressed in ovarian tissue cancerated. It's expression more fell after chemotherapic drug-resistance (PO.001, respectively); while the P-gp expression was opposite, up regulating after ovarian tissue cancerated, it's expression was more obvious in chemotherapic drug-resistance group(P<0.005, respectively). Conclusions: 1. In PEOC, low-level or even no expression of CX43 and up- expression of P-gp hint chemotherapic drug-resistance and prognostic badness; oppositely, higher expression of CX43 and lower expression of P-gp was suggest chemotherapic drug-sensitiveness and prognostic goodness.2. CX43 and P-gp are valuable indicator for the choice of clinical chemotherapic drug. In recurrent cases, chemotherapic resistance drug may be avoided by assaying CX43 and P-gp and so as to provide more effective treatment.
