| Objective vMIP- â…¡ was an analog of chemokine ,which has a broad-spectrumantagonism to other chemokine receptors, can bind CXCR4.Thus vMIP- â…¡ affected the biological function of chemokine receptor CXCR4.. The receptor CXCR4 main located on the surface of cells.The aim of this study was to make certain role of vMIP- â…¡ in CXCR4 internalization,which can explain the basic mechanism of vMIP-â…¡ in the role of anti-HIV/AIDS.Methods Here laser scanning confocal microscope was used to determine thenature of CXCR4 internalization.Flow cytometer was used to quantify CXCR4 internalization.We also applied spectrofluorophotometor continuous to examine dynamic characteristics of Ca2+ influxion.Results Laser scanning confocal microscope showed that vMIP- â…¡ can induceCXCR4 internalization. Flow cytometer showed that 100ng/ml vMIP-â…¡ can induce 75% CXCR4 internalization,what more important is the intemalizated CXCR4 can not recycle to the cell surface.We applied spectrofluorophotometor and found vMIP-â…¡ can induce Ca2+ afflux quickly,also vMIP- â…¡ can prompt Ca2+ release from Ca2+ reservoir.Conclusions vMIP- â…¡ can induce CXCR4 internalization,the max value is75%,also the intemalizated CXCR4 can not recycle to the cell surface.Thus vMIP- â…¡ reduced the quantity of CXCR4 located on the cell surface. vMIP- â…¡ can induce Ca2+ influx quickly,it can also prompt Ca2+ release from Ca2+ reservoir.So,vMIP- â…¡ is a quite useful protein to anti-HIV/AIDS. |
