Cloned Cytochrome Oxidase Subunit Ⅰ (CO Ⅰ) Gene And Internal Transcribed Spacer 2(ITS2) Gene Of Paragonimus Westermani For Species Identification

Objective: To evaluate paragonimus westermani COâ… and ITS2 genesfor species identification and for studying parasite populations. Methods:DNA from an adult fluke of Paragonimus westermani, Pargonimusskrjabini, and Euparagonimus cenocopiosus was extracted. DNAfragments of CO â… and ITS2 from Paragonimus westermani wereamplified,cloned, and sequenced. The genes were labeled with digoxigeninand used as probes. DNAs from the various parasites and rabbit controlwere applied to nylon transfer membranes, hybridized with labeled probes,and visualized according to the manufacturer's instructions. Nucleotidesequences were calculated by DANMAN.Result: Nucleotide sequences of COâ… and ITS2 from geographicalisolate of paragonimus westermani from china, Japan, Korea, Taiwanwere calculated by DNAMAN. The paragonimus westermani in researchwas recognized in group of northeast Asia which was relatively uniform.The CO â… probe hybridized well with DNA from Paragonimuswestermani, but not with DNAs from other species, e.g. Paragonimuswestermani specific. In contrast, ITS2 could hybridize with DNA fromboth Paragonimu westermani and rabbit.Conclusions: The paragonimus westermani in research was recognized ingroup of north east Asia which was relatively uniform .The COâ… gene ofthe paragonimus westermani can used for species identification.
