Objective:To investigate the expression of angiopoietin2 gene in glioma and itsrelationship with angiogenesis.Methods:Expression of angiopoietin2 in 52 human and 8 normal brain tissues wereexamined by RT-PCR and SABC, The vascular development was measured bymicro-blood-counting (MVC) which was immunostained with anti-CD34monoclonal antibody .Resurlts:Expression of ang2 transcript was found .in 50 human gliomas and 8 normalbrain tissues, expression of ang2 mRNA was significantly positive correlated withMVC, (r=0. 821,P<0 01), among which the expression of ang2 mRNA andprotein level ,MVC separately in malignant glioma ware considerably higher thanthose in low grade glioma(P<0 01); The ang2 mRNA and protein level ,MVC inlow grade glioma were significantly higher than those in normal brain tissues(P<005). Immunohistochemical staining showed the distribution of ang2 protein wasmainly in tumor cells and endothelial cells of malignant glioma,whereras it was onlyweakly expressed in low grade glioma and it wasn't expressed in normal braintissues. Conclusion : Expression of ang2 was significantly positive correlated with pathological gradein glioma; Ang2 may contributed the progression of human glioma by playing animportant role in leading to angiogenesis.
