OBJECTIVE:1. To explore the best method of fundus fluorescein angiography in rats, and determine the suitable dosages of fluorescein and indocyanine.2. To set up retinal and choroidal fluorescein angiography parameters in rats and offer reference to living observation of retina and choroid.3. To offer normal data for experimental study model of vascular diseases in rats.METHOD:1. Blood vessel of retina and choroid was observed by confocal laser scan instrument for angiography with fluorescein and indocyanine green in rats.2. 9 healthy and mature Wistar rats were randomly divided into three groups, and mixed contrast solutions of different dosages of sodium fluorescein and indocyanine green were given through caudal vein respectively and blood vessel of retina and choroid was observed by confocallaser scan instrument. The best dosages was determined in accordance with the best resolution of pictures selected by double-masked method.3.14 Wistar rats were used to determine the vascular characters of retina and choroid with the method described above and the best dosage determined above.4. 15 Wistar rats were divided into three groups randomly, and mixed contrast solutions with different dosages of sodium fluorescein and indocyanine green were injected intraperitonealy and angiogram of retina and choroid was captured with the method described above. The best dosage of angiography by peritoneal injection was also determined with the method decribed above.5. All processes of angiography were lasted about 40 minutes. RESULT:1. Suitable dosage of fluorescein for FFA via tail vein is 40mg/kg(10% sodium fluorescein), while 6mg/kg(1.25% indocyanine green) for ICGA. Suitable dosage of fluorescein for FFA by peritoneal injection is 120mg/kg(10% sodium fluorescein),while no for ICGA.2. Mean time of tail-retina circulation time of fluorescein is 6.6 +3.0s, mean time of retinal vein filling time is 9.3 +3.5s, mean time of retina circulation time is 2.1±0.58s,and mean time of fade-out time is 18.1 + 5.1min.3. Mean time of tail-choroid circulation time of indocyanine green is 6.6 + 3.0s, mean time of choroid artery and vein filling time is 11.5 + 2.1s; mean time of fade-out time is 15.3 ±5.1min.4. Six to eight retinal branch arteries with the same number of retinalbranch veins were observed from optic disc in radial order.5. Many vascular connections between retinal shallow layer capillary network and deep one were observed via vertical blood vessel.6. The choroidial arteries locating in inferior temporal part of optic disk look like Straight-line, comb-shaped choroidial little veins can be seen converging to ampulla-like vortex vein. Mean time of venous-artery filing time is 40.9+4.80s and mean time of flourescein fade-out time is 20.4 ± 2.70min by peritoneal injection.7. Clear picture of retinal blood vessel was seen using FFA by peritoneal injection, while not the retinal and choroidal blood vessel using ICGA. CONCLUSION:Fundus angiography system can be used to observe retinal and choroidal blood vessel in rats. Suitable dosages of dyes, correct routine of dye injection and continual picture capture are necessary for obtaining retinal and choroidal angiography. Simultaneous fluorescein and indocyanine green angiography is one of the best observation methods for experimental study model of retinal and choroidial vascular diseases in rats.
