| As Traditional Chinese Medicine (TCM) of tonic function, Radix astragali has been used thousands of years with folk foundation. One of strategic target of TCM modernization is to provide TCM with the standard criterion accepted by the international, only with the modern evaluation system of TCM quality has been constitute and consummate. The quality of TCM can be evaluated by the chromatographic fingerprints, which represent the compound groups of TCM and substantial foundation of pharmacodynamics. The chromatographic fingerprints can cater to the theory of Chinese traditional medicines and characteristics of chemical constituents in TCM, thereby also reflect the quality of TCM in an all-around way and be accepted by international. The article established the quality control methods of Radix astragali with HPLC/DAD, HPLC/ELSD and HPLC/MSD, the relative research of chromatographic fingerprints had been performed. I. HPLC/DAD chromatographic fingerprints of Radix astragaliFirstly, the pretreatment conditions of Radix astragali were inspected, the solvent was chosen by the extraction method of soak and soxhlet, the most optimized condition of ultrasonic was ascertained by orthogonal design, and ultrasonic extraction with methanol twice, 20 minutes each time was thought to be the best. Calycosin-7-O-β-D-glucoside and formononetin in Radix astragali were determined by HPLC/DAD. Secondly, the HPLC/DAD chromatographic fingerprints of Radix astragali from ten different batches were established, the methodology was inspected and eleven common peaks were affirmed, the standard chromatographic fingerprint was established and the similarity was calculated. With the inspection of similarity at different wavelengths, the similarity evaluation method that assembled the characteristic information of all common peaks at different wavelengths was brought forward. The relative parameters of chromatogram were evaluated by chemometrics, 260nm was thought to be the optimized detection wavelengths through the discussion.
