Objective To study the expression of BPl gene in breast cancer and to investigate the relationship between BPl gene and the clinicopathological factors. Methods The expression of BP1 gene mRNA in 52 cases of fresh breast cancer and 16 cases of fresh normal breast tissue were detected by reverse transcriptase porymerase chain reaction (RT-PCR). The TUNEL-positive cells were counted as apoptosis index(AI). Using in situ hybridization method, the expression of BPl gene mRNA was investigated in paraffin sections of 165 breast cancer samples. The expression of ER, C-erbB-2, p53, PCNA, bcl-2 of the two groups of breast cancer were detected by immunohistochemistry. Results There was a statistical difference of BPl expression between breast cancer and normal breast tissue(P < 0.01). A significant relationship was observed between the expression of BPl and the status of ER, bcl-2, (P < 0.05) and AI (P < 0.01) in breast cancer by RT-PCR. By in situ hybridization method, a significant relationship was observed between the expression of BPl and the statue of bcl-2 (P < 0.01) and both of them displayed a closed correlation with ER (P < 0.05) and p53 (P < 0.05) .Conversely, closed relationship was not discovered between the expression of BPl and the status of C-erbB-2, PCNA, age, tumor's size, histological type, lymph node metastasis and histological grade. Conclusion BPl may play an important role in breast cancer through inhibiting apoptosis and serve as a marker for prognosis and therapy.
