OBJECITIVES: The purpose of this experiment is to investigate the antioxidative activity of leaf protein concentration extracted from alfalfa by heat coagulation , based on the analysis of the GSH-Px activity , SOD activity and MDA content,which are in the blood of normal adult mice , in the blood and liver of adult mice who were exposed to 6.5Gy dosage 60Coγ. METHODS:The ALP used was concentrated by heat coagulation .The female NIH adult mice were randomly divided into 6 groups according to their body weight,each 23 animals with 22±3g. I: High-dosage group;II: Middle-dosage; III: Low-dosage group; IV: Postive control group; V: Model control group; VI:Normal control group。The mice were fed for 40 days ,then exposed to 6.5Gy dosage 60Coγ in order to obtained Lipid peroxidation model. Three days after exposing to 60Coγ,all the mice are scarified and the livers were removed. The antioxidative activities of ALP were appreciated by determining the GSH-Px activity ,SOD activity and MDA content,which are in the blood of normal adult mice and in the liver of adult mice who were exposed to 6.5Gy dosage 60Coγ;The effects on growth and development of normal adult mice were assessed by hemoglobin and body weight in the different experimental course. RESULTS: 1. The ALP concentrated by heat coagulation contained plenty of protein and amino-acids. The amino-acids composition is in proportion. 2. Administration of ALP leads to the marked increase in activity of GSH-Px and SOD and the significant decrease of MDA content in blood of normal adult mice(P<0.05). 3. The results showed ALP would prevent decrease of live GSH-Px activity and increase of liver MDA content in adult mice, which were induced by radiation of 6.5Gy dosage 60Coγ(P<0.05). 4. ALP wouldn't increase Hb content in blood of normal adult mice ,but prevent body weight of normal adult mice from increase(P<0.05). CONCLUSION: 1. ALP would play a role in improving antioxidative ability of body,decreasing the damage of free radical to the body and maintaining the body weight of normal adult mice.The antioxidation of ALP maybe relate to Se,Vitamin E,β- carotin and flavonoids that ALP contained. 2.ALP has the different antioxidative mechanism from that Vitamin E has. 3.However ALP is inferior to Vitamin E in decreasing MDA content inthe liver of radiated adult mice ALP ,it is a kind of healthy food for its perfect Aa compostion and function to maintain the adult mice body weight.
