| Objective: To study the curative effects of rutongning capsule on mammary gland hyperplasia in rats and provide the basis for the wider use. Methods: Forty-five super Wistar mature female unpregnant rats were randomly divided into five groups: the normal control (five), the model control (ten), the treatment group by lower dose (ten), the treatment group by middle dose (ten), the treatment group by higher dose (ten). Except for the normal group, all the other four groups were injected with estradiol (0.5mg/kg) once a day for twenty-five days. Then the four groups were injected with progesterone (5mg/kg) once a day for five days to induce mammary gland hyperplasia models. Then, except for the normal group and the model group, all the other three groups were perfused with rutongning once a day for thirty days, the lower dose is 70mg/kg, the normal dose is 140 mg/kg, the higher dose is 280 mg/kg. Then all the rats were killed and serum were separated from blood. Total estradiol (E2), prolactin (PRL), follicle stimulating hormone (FSH), luteinizing hormone (LH), progesterone (PGN) and testosterone (T) in serum were determined by radioimmunity (RIA). The diameter of the second twin nipples was measured. Pathological histological examination was performed on the mammary glands. Estrogen and progesterone receptors (ER, PR) were determinded by immunohistochemical technique. Results: 1,The diameter of the second twin nipples by statistics and comparison show that the nipple diameter of the model group and the lower curative group is larger than the normal group (P<0.01) after the experiment. The model group and the lower curative group show no significant difference (P>0.05). The nipple diameter of the normal group and the middle curative group and the higher curative group shows no significant difference (P>0.05). The nipple diameter of the normal group and the middle curative group and the higher curative group is much smaller than the model group (P<0.01). 2,The hormone in serum of all the rats by statistics shows that PRL,FSH and E2 in model group is much higher than the normal group (P<0.05 or P<0.01). LH and PGN in model group is much lower than the normal group (P<0.05 or P<0.01). T in all rats is less than 0.5 ng/ml and can not be analysed by statistics. After the treatment by rutongning, PRL,FSH,E2 decreased and LH,PGN increased. But the comparison between the lower curative group and the normal group shows significant difference (P<0.01). The lower curative group and the model group show no significant difference (P>0.05). The comparison between the middle curative group,the higher curative group and the normal group shows no significant difference (P>0.05). The comparison between the middle curative group,the higher curative group and the model group shows significant difference (P<0.05 orP<0.01). 3,Pathological histological observation proves that the rats in normal group have no mammary gland hyperplasia. And all the rats in model group have mammary gland hyperplasia. We find that mammary gland hyperplasia in rats in model group diffuses, more acini and small lobes, less fat and connective tissue, enlarged acinus cavity and gland duct. Compared with the model group, the treatment group shows much less acini and lobes after the treatment by rutongning. The acini atrophy, the ducts and volume become small. Some can come back to be normal. The middle dose group and higher dose group show similar improvement and so significant. 4,The number of estrogen and progesterone receptors (ER, PR): The number of ER and PR positive cells in model group is much more than that of normal group. The curative groups show a few of positive cells. The middle dose and higher dose group are much better than the lower dose group. Conclusion: 1,Rutongning capsule can rectify the secretion of hormone about the pituitary-sex gland axis in rats that have mammary gland hyperplasia and adjust the hormone level in rats that have hormone disorder. Rutongning capsule can decrease PRL,FSH,E2 and increase LH,PGN, so it can cure MGH. 2,Rutongning capsule can decrease the diameter of the second twin nipples in rats that have MGH . Rutongning capsule can make the acini atrophy, and make the gland ducts and volume become small. Many rats can restore to health. |
