2DM (2 diabetes mellitus) is a kind of endocrinopathy andmetabolized disease which influence a lot of organs and systems. Itcan lead to some serious complications such as blood vessels, capillary vessels and so on. Some researches indicated that there arenotability relations between diabetes and dementia especiallyAlzheimer's disease. IDE had been shown to degraded a number ofbiologically important peptides, including insulin and Amyloid-βprotein which is implicated in Alzheimer's disease. Thereby IDEcould be important in the reasons of diabetes and its complication.IDE is a 110KD neutral metalloendopeptidase and its cDNA isabout 3kb which own two initiation codons. It has some othersubstrates which including glucagons, insulin-like growth factor Ⅰand Ⅱ, atrial natriuretic factor, transforming growth factor-α, growthhormone releasing factor and β-endorphin.Generating enough IDE with a kind of simple method is notableto research the structure and function of the IDE. In 2000, a researcherexpressed the IDE in the E.coli the first time in the Chicago University,USA and made the human recombine IDE from the second initiationcodon. The IDE was cloned by the pcDNA/rIDE as template fromHarvard University and subcloned to the plasmid pGEX. Thesequence was test and the result is same to the data published on the...
