Study On Alkaloids From Macleaya Cordata â…¡

Macleaya cordata is Chinese traditional herbal drug with plenty of alkaloid, which has pharmacologic function of antibacterial action and so on. In this paper, it was summarized briefly that Macleaya cordata, Papaveraceae, the application of ultrasonic technology and macroporous resins on adsorption and separation of chemical constituents from Chinese herb medicines. The extraction technology and alkaloids from Macleaya cordata were studied. The main results as following: The chemical constituents in the Macleaya cordata have been pretested systematically by means of tube-test method. It was found that plenty of alkaloid, carbohydrate, phytosterol, terpenoid, flavone, phenylpropanoid and naphtha exist in Macleaya cordata. It does not contain anthraquinone, hydroxybenzene and amino acid, et al. The effects of alcohol concentration, liquid-material ratio and ultrasonic times on the extraction yields of alkaloids from Macleaya cordata were studied by use of L9(3)4　orthogonal test scheme, and the effects of adsorption and separation of alkaloids from raw extract with macroporous resins D101 were investigated. The results show that the optimal extraction conditions were A3B1C3,i.e. 9 times amount of 90% ethanol, ultrasonic extracting 45 min. All alkaloids in the hydrochloric acid solution of pH2-3 were abstracted in the form of free base, and that the polar alkaloids and weak polar alkaloids were separated by means of 50% ethanol elution and acetone respectively. With ultrasonic extraction, column chromatography and recrystallation, Seven alkaloid compounds were isolated from the Macleaya cordata . These compounds are as following: X-1(55 mg), X-2(70 mg), X-3(54 mg), X-4(22 mg), X-5(61 mg), X-6(127 mg) and X-7(104 mg). Based on chemical reactions and spectral analysis (UV, IR, ¹H NMR, ¹³C NMR, DEPT, EI MS and FAB MS), four of them were elucidated as 6-acetonyl-dihyrosanguinnarinc(X-2), berberrabine(X-4), dihydrochelerythrine(X-5) and α-allocryptopine(X-6) respectively. 6-Ace-tonyl-dihyrosanguinnarinc, berberrabine and dihydrochelerythrine were found from this plant for the first time.
