Objective To study the protective effects of Extracts from the Leaves of Ginkgo biloba on trichloroethylene induced human keratinocytes cytotoxicity.Methods Pool the keratinocytes derived from 3 or more mormal donors together and culture them with K-SFM, different concentrations of trichloroethylene were administered in logarithmic growth phase. Neutral Red Uptake Assay was used to determine the NR50, and the concentrations used in the following tests were determinated base on it. Different kits were used to measure the cells' LDH release level, MDA level, SOD level . ROS level and NO, NOS level. Transmission electron microscope and flow cytometer were used to determine the cell apoptosis level induced by trichloroethylene. At logarithmic growth phase, the cells were preincubated with different concentrations of GbE for 2 hours, then 2.0 mmol/L trichloroethylene and different concentrations of GbE were administered. Different kits were used to measure the cells' LDH release level, MDA level, SOD level and ROS level; Transmission electron microscope and flow ctyometer were used to determine the cell apoptosis level. The data was analyzed with SAS 6.12.Results The results included: (J) The NR50 of trichloroethylene on cultured human keratinocytes was 4.5252 mmol/L, with the 95% confidence interval of (3.92210, 5.12838); Positive control showed the NR50 of SLS on cultured human keratinocytes was 0.2773 mmol/L, with the 95% confidence interval of (0.19300, 0.36154).
