[Objective]:To observe the dynamic pathology of the posterior capsular opacification after ECCE, and to study the expression of matrix melloproteinase-2 (MMP-2) and its specific tissue inhibitor (TIMP-2) in the residual cells on it, so as to get the information that whether fibroblast cells and MMP-2/TIMP-2 have effects on the opacification of the posterior capsule.[Methods]:Twenty adult rabbits were studied. In each rabbit, one eye undergone extracapsuler cataract extraction and the other eye was normal control without any surgical intervention. The samples of the posterior capsular membrane were collected on the postoperative day 1, 3, 7, 14 and 28 respectively, and HE and immunohistochemical methods were employed to locating the fibroblast cells and the MMP-2/TIMP-2' expression on the posterior capsular membrane.[Results]: 1.HE method :There are no fibroblasts on normal lens. On the first day after ECCE , afew fibroblast cells were found attached to the posterior capasular membrane. On the third day, the amount of fibroblast cells increased obviously, and they appeared to be bilayered . Then, the fibroblasts were detected to be more and multilayered. 2.Immunohistochemical method:MMP-2 and TIMP-2 can be detected on the normal lens epithelial cells with a small amount. Their expression can be detected to be more . The longer the time after ECCE to be , the more expression of MMP-2/TIMP-2 will be found. [Conclusion]:1. On the early stage after extrcapasular cataract extraction , the fibroblast cells have effect on the opacification of the posterior capsule.2. MMP-2/TIMP-2 may be one of the major contributors to posterior capasular opacification and fibrosis.
