| As a beauty products, hair dyes are widely used in daily life.The components of the hair dyes include arylamine, polyhydric phenol,hydroquinone etc. This paper reports adverse effects of commercial oxidized hair dyes on mice and human such as subchronic toxicity and mutagenicity .Part one: Commercial oxidized hair dyes were applied to the skin of mice once a day for a month. During this period, the biological effects of the dyes were observed including body weight, biochemical examination of blood, histopathology of skin, liver, spleen, lung and kidney, and bone marrow genotoxicity. The results are as follows: as the dose increased, the blood biochemical indicators(ALT, ALP, BUN and CR) changed significantly, the weight of body and organs( liver, lung )decreased but that of kidney increased . Alteration of histopathology in high dose group displayed many nuclei pyknosis in germinal cells and sebaceous gland cells.The germinal cells peri-hair follicle decreased and inflammation infiltrated in coriun tissue . Compared to the control and low dose group, the hair follicle decreased significantly in high does group. The DNA strand breaks could be observed in the mouse bone marrow cells and epidermic cells.Part two: To study potential health effect of hair dyes such as DNA damage and apoptosis, the technique of Single Cell Gel Electrophoresis(SCGE) was used and ten people were exposed to the hair dyes, the DNA damage of hair follicle after 3h, 7h, 24h of exposure significantly increased, compared to the control group. The AO/PI method and Laser Confocal Fluorescence Microscope(LCFM) were used and revealed a higher rate of apoptosis and cell death of the hair follicle.The results indicated that the hair dyes could damage the DNA of mice skin and human hair follicle cells, and the liver and kidney functions of miceâ– > The experiment provided with the evidence of genetic toxicity of oxidized hair dyes on mammals,... |
