Endometriosis is a common and popular gynecologic disease in women of the reproductive age. Recently, the incidence rate of endometriosis has been increasing that aroused many experts attention. Many evidences have demonstrated that the occurrence of endometriosis is a complicated process in which many factors participate, which is the primary reason that influences the therapeutic effect of this disease. It mainly includes three stages: adhesive, angiogenesis and implantation. Hepatocyte growth factor (HGF) and its receptor, C-Met, are likely to be involved in this process and they can increase the protein expression of vascular endothelial growth factor (VEGF) gene, which involved in the pathogenesis and development of endometriosis and had important role in the process of angiogenesis. We used immunohistochemical S-P methods to detect the protein expression status of HGF, its receptor C-Met and VEGF in a series of endometriosis and control endometrium in order to explore the relationship among this three factors and find a valuable target for early diagnosis and therapy of endometriosis and provide a new method for effectual remedy to endometriosis.Material and MethodPatients with endometriosis (n=47) acceptted operation and the sepciman were collected at gynecological department and pathologic department of the Third Affiliated Hospital of ZhengZhou University. The age of patients ranged from 29 to49. In addition, 35 control endometriums were collected in the same hospital, the age ranged from 30 to 51. All patients had not physical and surgical disease and had not accepted the hormone therapy six months before operation. All diagnoses of specimens were confirmed by pathology.In this study, we had explored the protein expression status of HGF> C-Met and VEGF in endometriosis and their clinic pathological associations in endometrium tissues by the method of immunohistochemical.SPSS12.0 was performed to compare the data, and a value =0.05 was taken as statistical significance.Result1. The positive rate of HGF protein is 17.14% in control endometrium, 78.72% in eutopic endometrium and 74.47% in ectopic endometrium. The positive rate in eutopic and ectopic endometrium group was higher than that in cortrol endometrium group (P<0.05). There is no signicificant difference between eutopic and ectopic endometrium group (P>0.05). The expression of HGF protein shows regularly in control endometrium, which is higher in the secretive phase than that in the proliferative phase. There is not this phenomenon in eutopic endometrium of endometriosis, and the expression is higher in secretive and proliferative phase than that in the control group.2. In eutopic endometrium group, the positive rate of C-Met protein is 70.21% and in ectopic endometrium group is 76.60%. There is no significant difference between these two groups (F>0.05), whereas, in control endometrium group is 20.00%. The positive rate of endometriosis was significantly higher than that of control endometrium group (P<0.05). The expression of C-Met is regular in control group, which is higher in the secretive phase than that in the proliferative phase. But we did not find this phenomenon in eutopic endometrium of endometriosis, and the expression is higher in whatever phase.3. In control endometrium, eutopic and ectopic endometrium group, the positive rate of VEGF is 22.86%, 74.47%, 80.85%, respectively. There was obvious differencebetween eutopic and control endometrium group and between ectopic and control endometrium group (i*<0.05). There is no statistical significance between eutopic and ectopic endometrium group (P>0.05). The expression of VEGF protein shows regularly in control endometrium, which is higher in the secretive phase than that in the proliferative phase. There is not this phenomenon in eutopic endometrium of endometriosis, and the expression is high in secretive and proliferative phase.4. In eutopic endometrium group, there were 33 cases which had both HGF and VEGF expression at the same time, 8 cases had negative expression. Whereas, 34 cases had positive expression of both HGF and VEGF, and 8 cases had negative expression in ectopic endometrium group. According to the relative analysis, thecorrelation coefficient rs =0.649, i><0.05 and rs =0.707, P<0.05, respectively.Conclusions1. HGF and C-Met expression in eutopic and ectopic endometrium group was significantly higher than that in control endometrium group, and if s expression was not obviously associated with phase. It indicates that HGF and C-Met have an important role in endometriosis development and progression.2. The expression rate of VEGF in eutopic and ectopic endometrium was higher than that in control endometrium. It suggests VEGF play a principal status in the development of endometriosis.3. The expression of HGF has a positive correlation with the expression of VEGF, which suggest that they can promote the development and progression of endometriosis together. Therefore, they may become the new factor to the early diagnosis and therapy of endometriosis.
