Objective: To study the expression of VEGF, p53, MDM2 proteins and their relationships in patients of hepatocellular carcinoma (HCC) and analyse their Clinical Significance.Methods: VEGF, MDM2, and p53 proteins were detected by immunohistochemical technique with SP or SAP, and the tissue microvascular density (MVD) was measused in 72 cases of HCC. t 25 cases of paracancerous tissues and 10 cases hepatic tissues were selected as normal controls.Results: The positive rates of VEGF, MDM2 and p53 proteins in HCC tissues were 62.5%( 45/72 ), 41.6%( 30/72 ) and 33.3%( 24/72 ), respectively. Expression of VEGF was positively correlated with expression of p53 ( P<0.05 ) and MDM2 ( P<0.05 ). The frequencies of VEGF, MDM2 and p53 protein positively in corresponding paracancerous tissues were 20.0% ( 5/25 ), 8.0% ( 2/25 ) and 8.0%( 2/25 ), respectively. There was a significant difference in VEGF, MDM2 and p53 protein expression between HCC and paracancerous tissues (P<0.05 ). Expression of VEGF, MDM2 and p53 was not detected in normal hepatic tissue. Expression of VEGF, MDM2 and p53 and higher MVD showed a significantly positive correlation with vascular invasion and metastasis. According to Cox's hazards analysis, univariate and multivariate analysis showedvascular invasion were related to the reducing of survival. Conclusion: There is a high expression rate of VEGF, MDM2 and p53 proteins in HCC by immunohistochemical technique detecting. The expression of VEGF, MDM2 and p53 protein is related to vascular invasion and metastasis in HCC. They may be used as the useful predictor of poor prognosis in patients with HCC.
