Objective:To study the expression of hypoxia inducible factor1α(HIF-1α) and vascular endothelial growth factor ( VEGF ) correlate withangiogenesis in renal cell carcinoma(RCC)Methods: 50 specimens of our hospital renal cell carcinoma and 10specimens of our hospital normal renal tissues were selected,men 33case,women 17 case,age:17-75 years old,average 56.7years old.paraffin-embedded sections of 50 cases of renal cell carcinoma and 10cases of control kidney were immunohistochemical staining for HIF-1αandVEGF using a standard streptavidin-biotin complex procedure for lightmicroscopic investigation. Microvessel density(MVD)was assessed byimmunohistochemical staining for endothelial cells using anti-CD34monoclonal antibody. tumour grade: Robson stagingⅠ14 case, Robsonstaging Ⅱ25 case, Robson staging Ⅲ7 case, Robson staging Ⅳ期4 case;Fuhrman staging G1 12case, Fuhrman staging G2 19case, Fuhrman staging G314case, Fuhrman staging G4 5case,Results: In 50 specimens of renal cell carcinoma patients,the positiverates of HIF-1αprotein were higher in renal cell carcinoma (54%,27/50)than in normal renal tissues(0%), There was the significanct diferencebetween HIF-1αprotein expression rates of renal cell carcinoma andnormal renal tissues (P<0.01). The positive rates of VEGF protein werehigher in renal cell carcinoma 64%(32/50) than in normal renaltissues(20%,2/10), There was the marked diference between VEGF proteinexpression rates of renal cell carcinoma and normal renal tissues (P<0.05).The positive rates of MVD were higher in renal cell carcinoma (57.59±8.47) than in normal renal tissues(21.32±7.64%), There was thesignificanct diference between MVD expression rates of renal cellcarcinoma and normal renal tissues (P<0.01). The MVD expression werehigher in the positive group of HIF-1αprotein than in the negativegroup of HIF-1αprotein, there was a positive correlation between HIF-1αprotein and MVD; The MVD expression were higher in the positive groupof VEGF protein than in the negative group of HIF-1αprotein, there wasa positive correlation between VEGF protein and MVD; there was asignificanct positive correlation between HIF-1αprotein and VEGFprotein。Conclusions:The positive rate of HIF-1αand VEGF gradually increased inrenal cell carcinoma,which may predict HIF-1αand its taget gene VEGFcanplay a very important role in the occurrence and development of renal cellcarcinoma. The expression of HIF-1αprotein were no correlated with thedegree of renal cell carcinoma. The expression of VEGF and MVD werecorrelated with the degree of renal cell carcinoma. The expression ofHIF-1αprotein were correlated with the expression of VEGF and MVD.Theexpression of HIF-1 αand VEGF protein accelerated not only theangiogenesis but also the development and incursions of tumor. Theoccurrence and development of renal cell carcinoma were the result ofpolygene's coordination.Through accelerated the angiogenesis of tumor,HIF-1αplay a important role in the development and incursions of renalcell carcinoma.So it may become a new target of anti-tumor therapy as amark of biologic behavior.
