| Background: Cholangiocarcinoma is a kind of malignant tumor which metastasize at the later stage. Cholangiocarcinoma have a high degree of malignancy and a poor prognosis. Cholangiocarcinoma is resistant to current surgical,radiotherapy and chemotherapy, therefore combining Chinese traditional medicine to cure cholangiocarcinoma is a virtually better method. Chinese traditional medicine Pingxiao capsule which had been proved by clinic is a curative effect very remarkable anti-Cholangiocarcinoma medicine. But academic world rarely report its research in pharmacology. Objective: The purpose of this study is to evaluate the growth-inhibiting and transfer-restraining effects of Pingxiao capsule on human cholangiocarcinoma cell line QBC939 cultured in vitro, find out the effect and aintcancer transfering mechanism of Pingxiao capsule on human cholangiocarcinoma cell line QBC939. Methods: By using the method of pharmacology of the serum with Chinese drugs, the Pingxiao capsule drug-serum was incubated with cultured QBC939 cells. The cell medicinal sensitivity,the cell cycle phase distribution and rate of apoptosis, the expression density of adhesive molecule CD44v6 on cell membrane as well as the viscoelastical properties of QBC939 cells and adhesive mechanical properties of QBC939 cells to CD44v6 monoclonal antibody were observed. Results: (1)MTT medicinal sensitivity method in vitro : Pingxiao capsule drug-serum could evidently inhibit the proliferation of QBC939 cells, which had obvious dose and time effect. (2)Flow cytometry analysis:After treating with the drug-serum, the proliferation of QBC939 cells was inhibited in G0/G1 phase. Pingxiao capsule could induce apoptosis, its rate of apoptosis increased with increasing of concentration of Pingxiao capsule. It could inhibit the expression of adhesive molecule CD44v6 on cell membrane, prevent QBC939 cells from transferring. Furthermore the expression density of CD44v6 on cell membrane decreased with increasing of concentration of Pingxiao capsule.(3)Micropipette aspiration methods:The effects of the viscoelastical properties of QBC939 cells were investigated from the view of Pingxiao capsule by micropipette technology. It was found that QBC939 cells presented a characteristic of high elastic coefficients and low viscous coefficients. After treating with the drug-serum, the viscoelastical properties of QBC939 cells-K1,K2,μevidently decreased, and K1,K2,μdecreased with increasing of concentration of Pingxiao capsule. After treating with the drug-serum, adhesive force of QBC939 cells to CD44v6 monoclonal antibody remarkably decreased, and adhesive force decreased with increasing of concentration of Pingxiao capsule, indicated the expression density of CD44v6 on cell membrane remarkably decline. Conclusion: Pingxiao capsule could evidently inhibit the proliferation of QBC939 cells, induce the apoptosis of QBC939 cells, inhibit the expression of adhesive molecule CD44v6 on cell membrane, and destroy cytoskeleton, consequently prevent QBC939 cells from transferring. |
