| Objective: To investigate the inhibitory effects of mizolastine on murine allergic contact dermatitis (ACD) and its mechanisms for scientific evidence of treating eczema and dermatitis with anti-histamines.Methods: Female BALB/C mice were chosen to establish the animal model of allergic contact dermatitis by the topical application of DNFB. The depressions on murine ear swelling responses treated with mizolastine before or after challenge were observed by detecting murine ear thickness and weight. The levels of IFN-γ, TNF-α and IL-4 in serum and suspension of cell-culture were detected by enzyme-linked immunosorbent assay (ELISA) in ACD mice treated with mizolastine.Results: Murine ear swellings were obviously depressed with each dose of mizolastine applied before challenge (P<0.05) and faded away earlier than those of control applied after challenge (P<0.01) . Murine ear weights of mizolastine groups had no difference compared with control (P>0.05) .The levels of IFN-γ in serum and the suspensions of spleen cell-culture as well as the levels of TNF-α in serum of ACD mice were significantly increased (P<0.01) , meanwhile the levels of TNF-α □from peritoneal macrophages and IL-4 in serum and the suspensions of spleen cell-culture were not changed ( P>0.05 ) . Mizolastine with different doses depressed these three cytokines evidently or in different degrees.Conclusions: Many cytokines are involved in the pathogenesis of ACD. Mizolastine is a kind of anti-histamine with anti-allergic and anti-inflammatory properties by inhibiting some inflammatory mediators and cytokines. In our experimental results, murine ear swelling responses were all depressed significantly treated with mizolastine before or after challenge. It demonstrates that mizolastine play its role on sensitization phase and effect phase. The levels of IFN-γ in serum and the suspensions of spleen cell-culture by ELISA wereincreased but not IL-4 in ACD mice. It indicates that Th1 cell play a key role in the immunity reponse of ACD. TNF-α was also involved in the pathogenesis of ACD. Furthermore, our results demonstrate that the depressive effects of mizolastine on murine ACD are exerted probably by inhibiting some cytokines, and it provides scientific evidence for the treatment of eczema and dermatitis with anti-histamines. |
