Vascular endothelial growth factor is a potent inducer of angiogenesis which was identified years ago, and a term of "therapeutic angiogenesis" has been proposed based on its key role in the pathologic process following cerebral ischemia. Although some clinical trails have shown that Chuanxiong and its compounds are effective in the treatment of ischemic cerebraovascular disease, it is unclear whether they have any effect related to therapeutic angiogenesis or not. Objective:To study the effect of Da ChuanXiong Pill—a kind of compound on vascular endothelial growth factor secretion by human umbilical vein endothelial cells cultured in vitro and find out if it has any effect ralated to therapeutic angiogenesis, so as to provide laboratory evidence for it's clinical usage in treatment for ischemic diseases. Method:Human umbilical vein endothelial cells were primary cultured andsubcultured in vitro with Jaffe method. According to the principle of seropharmacology of Chinese materia medica, cells were divided into high dose group, intermediate dose group and low dose group based on the drug concentration of serum added in, meanwhile, habbit-serum-absent control group and normal habbit serum control group were set. Each group was diveded into two equal parts to be cultured in vitro under the hypoxia and normoxia condition respectively, with contained-herb serum of three different drug doses added into culture media correspondently, and the concentrations of vascular endothelial growth factor(VEGF) in the supernatant were detected by enzyme-linked immunosorbent assay(ELISA). Result:The concentration of VEGF in habbit-serum-absent control group or normal habbit serum control group under hypoxia condition was significantly evaluated compared with that in corresponding group under normoxia condition(p<0.01). No significant difference of VEGF concentration was found between each experimental group and habbit-serum-absent control group under normoxia condition (p>0.05) ; the concentrations of VEGF were significantly increased in intermediate dose experimental group and high dose experimental group compared with that in habbit-serum-absent control group under hypoxia condition (p<0.01) . Conclusion:Blood vessel endothelial cells' secretion of vascular endothelial growth factor sharply increases under hypoxia condition, which is consistent with the conclusion of previous studies. VEGF secretion by human umbilical vein endothelial cells cultured in vitro can be effectively enhanced by the herb —contained habbit serum with specified herb concentration under hypoxia condition, but this do not happen under normoxia condition.
