Objective :To elucidate the effects of doxazosin on vascular stenosis after balloon injured abdominal artery on the hyperlipidemic atherosclerotic model of rabbits. And to find its relationship to serum lipid nitric oxide increasing and vascular smooth muscle cell proliferation in neointima and media.Methods: 23 New Zealand rabbits were randomlly divided into 3 groups.Normal contral group(n=5) was fed with ordinary diet and without any administration.The other two groups were fed with 1 % cholesterol diet , and their abdominal arteries were injured by dilated balloon.Meanwhile doxazosin group was fed with doxazosin controlled release tablet 4mg/d.Total cholesterol Triglyceride LDL—C HDL—C Nitric oxide were measured and they were compared with each other at 4th weeks after operating.Neointima thickness Media thinkness Adventitia thinkness Neointima area Lumen area Intimal elastic lamina and Extral elastic lamina were analyzed also by Hias-1000 imagining analysis system.PCNA positive cells in the intima and media were analyzed by half-quantity method using Hias-1000 imagining analysis system .In 3 groups vascular stenosis and PCNA positive cells were compared with each other after balloon injured.Results: The blood lipid in rabbit fed by high cholesterol diet were more higher than normal contral group.Vascular stenosis model were successed by using high cholesterol diet plus balloon injured method.There were no differense in blood lipid and nitric oxide at the begining of the test between doxazosin group and ballooninjure group(p>0.05).After operate for 4 weeks, the total cholesterol in doxazosin group was lower (p<0.05), and HDL-C was higher (p<0.05), nitric oxide was higher (p<0.001) compared with balloon injure group.The intima and adventitia thinkness were inhibited (all p<0.001) , neointima area were inhibited too (p<0.001) .Lumen area was increased (p<0.001), meanwhile intimal elastic lumina and extral elastic lamina were increased (all p<0.001) , PCNA positive cells in the intima and media was inhibited (p<0.001) .Conclusion :Doxazosin can inhibit vascular stenosis formed by using high cholesterol diet plus balloon injured method, can inhibit vascular smooth muscle cell proliferation, can reduce serum lipid and inhibite atherosclerotic progress, can increase serum nitric oxide.It may be helpful using doxazosin to diminish or prevent vascular restenosis after PCI, and this therapeutic methods will be useful to prevent vascular restenosis after PCI in clinc.
