| Objective In this study whether or not and how serum containing Gubenkechuangao induces proliferation and apoptosis of rat airway smooth muscle cells (ASMCs) cultured in vitro was observed, in order to explore the correlation between the asthmatic Sugen and airway remodeling, and clarify the reciprocity of asthma, and provide evidence in anti-recurrence of asthma in children together with the treatment of elimination and supplement.Methods Rat ASMCs were primarily cultured in DMEM containing 10% fetal bovine serum (FBS). Passage 4~7 of cells were used in the experiment. The effects of serum containing Gubenkechuangao on the growth of the cells and the cell proliferation models stimulated by IL-4 were detected by MTT method. Light microscopy was used to examine the morphological change. The apoptosis cell percentage was detected by in situ end labeling technique (TUNEL) of fragmental DNA.Results ①20μL serum containing high or middle dose Gubenkechuangao had the effects on inhibiting the proliferation of ASMCs normally cultured (P<0.05, P<0.01). ②Compared with the control group, IL-4 obviously induced the proliferation of ASMCs (PO.01). The OD values of ASMCs stimulated by IL-4 together with serum containing high or middle dose Gubenkechuangao, or with serum containing DXM, or with INF-γ were respectively lower than those of IL-4 group or IL-4 together with serum without medicine group (P<0.05), but there is no significant difference between the former groups. ③Light microscopic examination showed cell shrinkage,membrane blebbing, nuclear contraction, chromatin condensation and nuclearfragmentation in rat ASMCs. @Apoptotic index of rat ASMCs in serum containinghigh dose Gubenkechuangao group was significantly higher than in the control groupor serum without medicine group (PO.01).Conclusion Gubenkechuangao has the effects on inhibiting the proliferation of ratASMCs cultured in vitro. By inhibiting the promotion effect of IL-4 on rat ASMCsproliferation and inducing apoptosis of them, it can intervene the occurrence anddevelopment of airway remodeling, which shows the reciprocity of asthma with acertain degree. |
