Objective To study the programmed cell death of human uterine cervical cancer induced by arsenic trioxide and investigate the arsenic trioxide significance of clinical application. Method MTT reduction assay was adopted to detect the proliferation activity of HeLa cells which were treated by As2O3 of 0,1,2.0, 5.0, 10.0μmol/L in 24,48,72,96 hours respectively; Morphology investigation with HE and inverted phase contrast microscope was adopted ; apoptosis was detected by TUNEL and fluorescent staining(AO/EB); technology of immunohistochemistry was applyed to detect proteinum express of bcl-2/bax gene which were correlated to apoptosis. Flow Cytometry applyed to analysis cell cycle. Results The survival of HeLa cell line was inhibited by As2O3 proliferation activity of HeLa cells treated with AS2O3 of 2.0μmol/L decreased markedly(p<0.05) in 24 hours,and in 72 hours this condition (p<0.01) was better than it in 24 hours; a typical apoptotic morphology (volume of cells deflated,chromatin pyknosis,nuclear fragmentation, apoptotic body )also can be seen during the cells treated with AS2O3; Moreover DNA fragmentation can be detected by TUNEL ; the express of bcl-2 proteinum decreased but express of bax increased; Treated by AS2O3 of 2.0, 5.0, 10.0μmol/L, Flow Cytometry showed that ceil numbers of G2/M phrase was 3.6%,4.0% and 4.5% at 24 hours, and 3.8%, 7.9%and 10.5% at 48 hours, respectively. Conclusion AS2O3 can induce programmed cell death of HeLa cells and inhibit the cellular proliferation. The effect shows time and concentration dependence. Cell cycle changed markedly in Flow Cytometry. G2/M phrase increased but Gl phrase decreased. It indicated that HeLa cells treated by AS2O3 could be blocked in G2/M phrase. Mechanisms of apoptosis and cell cycle arrest of HeLa cells induced by AS2O3 have important significance in cervical cancer treatment.
