Objective The purpose of this research is to investigate the expression of a novel inhibitor gene of apoptosis, Surviving, during establishing multidrug-resistant human hepatocellular carcinoma cell line (Bel-7402/Dox) ,explore the relationship between expression of Survivin and multidrug resistance in an multidrug-resistant human hepatocellular carcinoma cell line.Methods Using gradually increased concentration of doxorubicin in culture, an doxorubicin-resistant human hepatocellular carcinoma cell line (Bel-7402/Dox) was established in vitro. During the progress, the rate of restrain was evaluated by MTT assay, the apoptosis rates of the hepatocellular carcinoma cell were measured by flow cytometer (FCM), and the change of expression of Survivin protein were detected by Western blot. Survivin mRNA of doxorubicin-resistant human hepatocellular carcinoma cell line (Bel-7402/Dox) was assessed by RT-PCR.Results With progressively increasing the concentration of doxorubicin (0.05μg/mk 0.1μg/mk 0.2μg/ml) , the apoptosis rates and the expression level of Survivin protein increased gradually. The expression of Survivin mRNA was positive in Bel-7402/Dox by RT-PCR.Conclusion The mechanism of Bel-7402 cell death induced by doxorubicin correlate with the apoptosis. The change of expression of Survivin might be correlated with the inquired drug-resistance of human hepatocellular carcinoma.
