Objective: Renal cell carcinoma is one of the most common malign tumor,and its morbidity accouts for 3 percent of totle sarcoma's one.what's more,its biological behavior is vary,and it is unclear for us to survey the biological basis of the mechanism,occurrence and development. Some recent studies survey that there is high expression of COX-2 protein and molecular level in cancer tissues and serial cell,such as enteron tumor,breast tumor,prostate tumor,bladder tumor,and so on.And the expression of Ki-67 associated with cell proliferation. To detect the expression of cyclooxygenase-2(COX-2) in human renal cell carcinomas,and its relationship to cellular proliferation.Methods: 45 RCC and 7 normal renal tissue were analyzed preoperatively for expression of COX-2 and Ki-67 by SP immunohischemistry (IHC) which was performed by using avidin-biotin peroxidase complex procedure.Results: (1) The positive rates of COX-2 expression in renal cell carcinoma were 23/45.But there was no and weak expression in noncancerous speciments of renal.Overexpression of COX-2 in renal cell carcinomas was related to its pathologic grading (F=22.463,P=0.000) ,clinical staging (F=13.215,P=0.001) and history type ( x~2=10.761, P=0.005); (2)Although expression of Ki-67 was found in all RCC,there was a faint stain in normal renal tissue.The expression of Ki-67 was closely related to the pathologic grading (F=30.715,P=0.000) ,clinical staging (F=18.497,P=0.005 ) and history type ( x~2=40.853,P=0.000) ; (3)The correlation was found between COX-2 and Ki-67 (Z=3.234,P=0.001) .Conclusion: The overexpression of COX-2 in renal cell carcinoma may play a crucial role in its pathologic grading,clinical staging, history type and cell proliferation in renal cell carcinoma.
