Objective 1. To observe the refrain role of proliferation on K-562 cells acted by matrine.2. To observe the differentiation role of matrine to K-562 cells.3. To find out the mechanism of Survivin gene in the process ofdifferentiation.Methods With varied concentration of matrine. using the cell culture technique. RT-PCR was employed to observe the expression of Survivin gene.Results In the certain range of concentration, Matrine can obviously refrain proliferation of K-562cells, induce cell differentiation and bring out alteration of cell morphology; After matrine was added to K-562 cells for a certain time, the expression of Survivin protein obviously decreased, and it had a significant difference between the trial group and the control group. Matrine can refrain cell growth with the dependence of time and concentration.Conclusion Matrine can strongly refrain growth of K-562cells. Survivin is a very important apoptosis regulation gene, and plays a role in the process of apoptosis of K-562 cells induced by matrine.
