Purpose: To observe the inhibitory effect of the 3-bromopyruvate on the cell proliferation of the human liver cancer SMMC-7721 cell line, and explore the best concentration of the 3-bromopyruvate.Method: The human hepatoma cell SMMC-7721 was grown in the HDMEM culture medium supplemented with 10 % fetal bovine serum. The cells was treated with different concentrations of 3-bromopyruvate,and then cell growth was assessed using MTT assay, the change of SMMC-7721 cell was observed by convert light microscope, whether 3-bromopyruvate induces apoptotic or necrotic cell death was explored by Hoechst 33342/PI fluorescent staining and electron microscope. Result: (1) MTT test showed that the 3-bromopyruvate could suppress the cell proliferation of the human liver cancer SMMC-7721 cell line. The inhibitory effect was the most remarkable at the concentration of 0.75 mmol/1. (2) the increasing necrotic cells had been observed by fluorescent staining, convert light microscope and electron microscope. The apoptosis was not remarkable. Conclusion: 3-Bromopyruvate could suppress the proliferation of the SMMC-7721 cell at the concentration 0.75 mmol/1.Purpose: To observe the inhibitory effect of the 3-bromopyruvate on the mouse tumor H22 by three giving-drug ways, and explore primarily the mechanism. Method: H22 cell was grown in the abdominal cavity of mice, on inoculation day, the tumor cells were harvested from abdominal cavity and washed with normal saline, then lxlO7/mL cells in 200 ul medium were injected into the groin of the ICR mice, after six days the mice were randomly divided into six groups, and the animals were treated with intraperitoneal, direct locally injection and gastro-gavaged the freshly prepared 3-Bromopyruvated 0.013 ml / g on separate days, and the control groups giving the same dosage PBS by the same way for seven days. Then weighed the mouse body and tumor nodules, calculated the size of necrotic cells of the tumor tissue in pathologic slice, measured the hexokinase activity of the tumor tissue, and examined the toxicity effects to the chief organs: liver, renal, heart and stomach. Result: (1) The mouse body weight haven't any changes, in contrast to control. (2) The tumor weight and HK activity were lower than control group giving drug by intraperitoneal and local injection. (3) The size of necrosis were bigger than control tissue giving drug by intraperitoneal and local injection. ( 4 ) 3-bromopyruvate haven't any side effect to liver, renal, heart and stomach. Conclusion: 3-Bromopyruvate could inhibit the growth of mouse hepatoma cell H22 in groups giving drug by intraperitoneal and local injection, 3-bromopyruvate haven't any toxicity to the chief organs, This research show that 3-bromopyruvate could suppress the proliferation of the hepatoma cell H22 through inhibiting HK activity.
