| Purpose To explore the serum protein mass spectrum of the carcinoma of endometrium to screen the differently expressed proteins and study their clinical diagnostic significance.Methods We selected 52 serum samples from patients with the carcinoma of endometrium and 44 serum samples from its control group.Using surface-enhanced laser desorption/ionization time-of-flight mass spectrometry technology (SELDI-TOF-MS) coupled with Weak-anion Exchange Chip(WCX2) we analyzed the proteomic spectrum of all serum samples,then selected randomly 30 serum samples from patients with the carcinoma of endometrium and 26 serum samples from its control group,searched the differently expressed proteins by Biomark Wizard Software,set up diagnostic cast of the carcinoma of endometrium by Biomark Pattern Software and obtained its positive predictive value and accuracy and specificity. Using this diagnostic cast we gave a double-blind trial to the remanent 22 serum samples from patients with the carcinoma of endometrium and 18 serum samples of its control group to get its accuracy and specificity.At last we researched the differently expressed protein on the UniProt protein database according to their molecular weight and isoelectric point.Results In the range of 0-50000Da,142 differently expressed protein biomarkers were found by the analysis of BioMark Wizard Software, among which 6 had statistical difference between the carcinoma of endometrium and its control group.2 biomarkers were low-regulated with molecular weight of 2748, 4106Da while 4 biomarkers were high-regulated with molecular weight of 4708, 8895, 9412, 18344Da in the serum samples of patients with endometrial carcinima. By the analysis of BioMark Pattern Software we set up diagnostic cast of the carcinoma of endometrium which comprising of 2 proteins with molecular weight of 2748, 9412Da and obtained its positive predictive value of 90.3% and accuracy of 93.3% and specificity of 88.5%.The diagnostic cast diagnosed all stage I samples correctly.Using this diagnostic cast we gave a double-blind trial to the remanent 22 serum samples of patients with the carcinoma of endometrium and 18 serum samples of its control group to get its accuracy of 81.8% and specificity of 83.3%.Searched inthe UniProt protein database, Histatin-3 l/24(Histatin-5) (pi: 9.82, MW: 2744.97) matches to the differently expressed protein of 2748Da.Histatin-5 is a potent inhibitor of MMP2 and MMP9.The latter can degradate extracell matrix to facilitate the development of neoplasm. No other proteins were found to match to another 5 differently expressed proteins in the UniProt protein database.Conclusion 6 differently expressed protein biomarkers were found between serum samples of patients with the carcinoma and those of its control group. The diagnostic cast of the carcinoma of endometrium which comprising of 2 proteins with molecular weight of 2748 n 9412Da not only obtained high positive predictive value ,accuracy and specificity,but also diagnosed all stage I samples correctly.So the diagnostic cast have significant value in diagnosis,especially early diagnosis of endometrial cancer. |
