| Objective To study the realationship between the urinary quantification of Isatin and the morbidity of malignant tumor. Methods The urine samples were extracted by acetic ether from 55 malignant tumor patients (24 thoracic cancer, 15 abdominal cancer and 15 urinary cancer) and 15 healthy controls. The quantification of Isatin was determined by HPLC: A ZORBAX SB-C18 colum(5μm,4.6mm×150mm) was used with mobile phase of acetonitrile-phosphate buffer(20:80, PH=5.8),The flow rate was 1 ml/min and the detection wavelength 242 nm. Besides isatin quantification in tumor group and control group, we compared isatin quantification in different cancer group, preoperational and postoperational isatin quantification of thoracic cancer patients, and isatin quantification in different pathological TNM stage group of thoracic cancer patients. Results Isatin had good linearity in 625-20000 ng/ml and its regression equation was C=0.00248X±52.82, r=0.9990. The RSD of inter- or intro-day was less than 8%.The average recovery was 99.66%. The average concentration of isatin is 2178.49±419. 25ng/mlCr in healthy controls, 2619. 77 ± 613. 64 ng/mlCr in thoracic cancer preoperationally, 2558. 99±535. 26 ng/mlCr in thoracic cancer postoperationally, 3914.29±1814.15 ng/mlCr in abdominal cancer, and 2963. 66 ± 634. 67ng/mlCr in urinary cancer respectively. The average concentration of isatin is 2664. 71 ± 718. 56 ng/mlCr, 2867. 02 ± 731.19 ng/mlCr and 3065. 39±816. 76 ng/mlCr in TNM stage â… ,â…¡ and â…¢ thoracic cancer patient respectively. The isatin quantification of tumor group, especially the abdominal cancer is significantly higher than that of control group (P<0.001) , TNM stage correlates with such change, but not significantly. After operation, the isatin quantification descended though not significantly. Conclusion The isatin concentration in urine correlates with the morbidity of malignant tumor. Periodic detection may show the morbidity, therapy effect, and prognosis of malignant tumor. |
