The technique of non-invasive diagnosis has been one of the research focuses in the field of biomedical engineering for a long time. Among which early diagnosis of lung cancer by detecting the volatile organic compounds (VOCs) in the exhalent breath is a quite promising technique.This work is based on some pilot study of the correlation of VOCs in the exhalent breath and many diseases. The SPME(solid phase microextraction) -GC(gas chromatography) system is used to detect and compare the VOCs in lung cancer patients' breath and in normal human's breath. An innovative method of lung cancer pathology analysis and early diagnosis on cell level has been introduced in this paper. The purpose of all these experiments is to find the correlation between lung cancer and the marker VOCs, and eventually certify the biomarkers in lung cancer patients' breath, and realize the non-invasive detection and early diagnosis of lung cancer by using the electronic nose we designed.There are mainly three parts in this thesis:First, some pilot study of lung cancer breath detection around the world is introduced and the comparison of routine lung cancer diagnosis method and e-nose breath detection method is made. Pathology analysis of the existence of special VOCs in lung cancer patients' breath and in lung cancer cells' metabolic product is introduced. The principles of SPME & GC and lung cancer culturing method which are closely related to our experiments are also particularly introduced.Second, detection and comparison of the VOCs in lung cancer patients' breath, in normal human's breath and in chronic bronchitis patients' breath are done with the SPME-GC system.Last and most important, the novel gas analysis system designed by us is used to detect the volatile organic compounds (VOCs) existing in the culture medium of ourseveral target cells after being cultured. After comparing the VOCs in culture medium of different kinds of lung cancer cells, lung cancer cells and normal lung cells, lung cancer cells and lung carcinoid cells, lung cancer cells and some other cells, special VOCs existing in the culture medium of lung cancer cells are found, which are the metabolic product of lung cancer cells and can be viewed as biomarkers of lung cancer. This research is significant to the development of breath diagnosis and non-invasive detection of lung cancer.
