Expression And Significance Of KAI1 And P53 In Gestational Trophoblastic Disease | | Posted on:2007-05-07 | Degree:Master | Type:Thesis | | Country:China | Candidate:Y Q Yang | Full Text:PDF | | GTID:2144360182494608 | Subject:Pathology and pathophysiology | | Abstract/Summary: | PDF Full Text Request | | Objective: To investigate the expression of KAIl, P53 protein, KAIl gene amplification and mRNA expression in gestational trophoblastic disease and the relationship between KAIl, P53 and its role in tumorgenesis, development and biological behaviour.Methods: The expression of KAIl protein, KAIl mRNA, P53 protein and KAIl gene amplification were investigated by immunohistochemical (SP method) and in situ hybridization in normal chorion of early gestation and tissues of gestational trophoblastic disease.Result: 1.The rate of KAIl protein expression in GTT was 17.4%(8/46) and the rate of KAIl protein expression in normal chorion of early gestation was 70%(14/20). The expression in normal chorion of early gestation was higher than that in GTT (P<0.05).2.The expression of KAIl protein was correlated to the age and tumor infiltration degree(P<0.05), while not correlated with the clinical stage, trophocyte proliferation and hyperplastic type of the patients (P>0.05).3.The rate of KAIlmRNA expression in GTT was 36.9%(17/46), the rate of KAIlmRNA expression in normal chorion of early gestation was 85%(17/20), Theexpression in normal chorion of early gestation was higher than that in GTT (P<0.05). 4. The expression of KAIlmRNA protein was correlated to the age and infiltration degree (P<0.05), while not related with the clinical stage, trophocyte proliferation and hyperplastic type of the patients (P>0.05).5. The rate of KAIl gene amplification in GTT was not higher than that in normal chorion of early gestation (P>0.05).6.The rate of P53 protein expression in GTT was 69.6%(32/46), The rate of P53 protein expression in normal chorion of early gestation was 10%(2/20). The expression in normal chorion of early gestation was lower than that in GTT (P<0.05).7.The expression of P53 protein was not correlated to the age, infiltration degree, clinical stage, trophocyte proliferation and hyperplastic type of the patients (P>0.05).8.The expression of KAIl protein was not found to be associated with the presence of P53 protein in gestational trophoblastic tumor (P>0.05,r=-0.S).Conclusions: l.The expression of KAIl is correlated to the progression of gestational trophoblastic disease. The patients with negative expression of KAIl are more easily evolve to GTT.2 With the deepening of infiltration, the expression of KAIl is reduced. It indicates that the reduction of the expression of KAIl is related to infiltration of GTT. This provides a useful index for evaluating metastatic potential and prognosis of GTT and provides some theoretical foundation and experimental evidence for further gene therapy.3.The expression of P53 is correlated to the progression of gestational trophoblastic disease.
