Aging is a globle problerm of 21 century,aged persons studied show asignificant decline in the immune response primarily caused by the immunesystem.Therefore, a lot of scientists devote their energy and efforts to theresearch on immnosenescence.At present,rigorous studies support the conceptthat aging is closely associated with an oxidative stress,the latter enhancesoxidative damage of protein and DNA,accelerate the process of decline ofimmune system,caused diseases.These studies offer new therapeuticapproaches toward immunosenescence of clinic, and provide new experimentprovement of new finding to pretest immunosenescence.D-galactose induced mouse aging model is a classic aging animal modeloften used in senescence studies in nearly 10 years. Because this model needless time and easier to success than normol aging mouse;and show similarchanges of 16-24 months age mouse.So it is used a lot in experiment ofresearch on aging.but the machnism of this model and the effect of VE pretectit from immnosenescence are not very clear ,these are the purpose of thispaper.The research programme is designed in two parts:part Ⅰ:theestablishment of D-galactose induced aging model;part Ⅱ:research if protecteffect of VE exist and the machnism of VE on aging induced by D-galactose.In part Ⅰexprement D-galactose was injected subcutaneously to establishaging model using KM mice.The result,which is detect ROS and otherimmune index, indicated that the model is successful.In partⅡ clystere these model with VE to prove that VE could protestaging rats to the damage of free radical,compared with control group ,ThymusIndex increased;the aptosise of thymus cells decreased;the proliferation of Tlymphocyte increased;the activity of IL-2 ,INF-γ,TNF increased and thekilling activity of non-specific killer cell increased.From these we can drow aconclusion that VE could protect aging rats to the damage of free radical andhelp to maintain the function of immune system.The detail is below.1.The establishment of D-galactose induced aging model80 Kunming rats were divided randonmly into 5 group.One group iscontrol group the others is modele group.Model group were injectedsubcutaneously with 40mg/kg.d.Forthermore dvided aging group with 4groups one is model control group.Other 3 group were clystered separatedwith 60mg/kg.d,30mg/kg.d,15mg/kg.d VE.After 10 weeks with continuousinjection and clyster. Caculate the Thymus index;obverve the aptosise ofthymus cells by FACS;MTT colorimetry was used to detect the proliferation ofT lymphocyte;check the activety of the cytocine IL-2,INF-γ and TNF bybiology activity method . Obverve the phagocytosis activity of Mφ fromabdominal cavity in rats by the test of englobemen neutral red;MTTcolorimetry was used to detect the killing activity of NK cell and CTL cell .Collected the blood specimens to detected the GLU,CHO,TG and superoxiderelative parameters SOD and MDA activity in the serum of these rats. Themodel group compared with control group, result is the Thymus Indexdcreased(P<0.05);the aptosise of thymus cells increased(P<0.05);theproliferation of T lymphocyte dcreased(P<0.05);and the activity of cytocineIL-2,INF-γ and TNF dcreased (P<0.05);the phagocytosis activity of Mφ fromabdominal cavity dcreased (P<0.05);and the activity of NK celldcreased(P<0.05);the GLU,CHO,TG increased obviously(P<0.05);theactivity of SOD dcreased obviously(P<0.05) and the activity of MDAincreased obviously(P<0.05).From these, it is significant that D-galactosecould increase the activity of ROS in aging model, and ROS maybe is the keyreason of immunosenescence of aging model.2. The interfereing effect on immunosenescence of VE in agingmodel rats induced by D-galactose :(1) The influence of VE on the function of T Cells of D-galactoseaging mice Modle:Caculate the Thymus index;obverve the aptosise of thymus cells byFACS;MTT colorimetry was used to detect the proliferation of Tlymphocyte;culture the cell with IMDM ConA,At last get out the upper leverafter 48 hours and checd the activety of the cytocine IL-2,INF-γ andTNF .Results:Succeede to get the D-gal aging mice model. Compared withmodel control group,the Thymus Index increased(P<0.05);the aptosise ofthymus cells decreased;the proliferation of T lymphocyteincreased(P<0.05);and the activity of cytocine IL-2,INF-γ and TNF increasedin the rats clysters by VE,but only 15mg/kg.d VE group shous appearlydiffirentily(P<0.05). From these we can drow a conclusion that VE couldprotect aging rats to immunosenescence;Enhance the activity of Tcells;Strenthen the function of immune systeme in aging rats.(2) The influnce of VE on specific and no-specific killer cells inD-galactose aging mice modleObverve the phagocytosis activity of Mφ from abdominal cavity in ratsby the test of englobemen neutral red;MTT colorimetry was used to detect thekilling activity of NK cell and CTL cell .Results:Compared with model controlgroup the phagocytosis activity of Mφ from abdominal cavity increased(P<0.05);and the activity of NK cell increased in the rats clysters by VE(P<0.05).But the killing activity of CTL cell didn't increase in the ratsclysters by VE (P>0.05). From these we can drow a conclusion that VE couldenhance the activity of special killer cells;but have no iflunce on the functionof special killer cells in aging rats.(3.) The influence of VE on biochemical parameters of D-galactose agingmice modle:Collected the blood specimens to detected the GLU,CHO,TG andsuperoxide relative parameters SOD and MDA activity in the serum of theserats.Results: Compared with control the GLU , CHO , TG increasedobviously(P<0.05) in aging rats .Cmpared with model control, the activity ofSOD increased obviously(P<0.05) and the activity of MDA dcreasedobviously(P<0.05) in aging rats were clysterd by VE. From these we can drowa conclusion that VE could protect aging rats to the damage of free radical;. Toprovide the experiment evidents of clinic use of VE to resist the aging.Above all,from the results of this paper :, The Thymus Index dcreased;theaptosise of thymus cells increased;the proliferation of T lymphocytedcreased;and the activity of cytocine IL-2,INF-γ and TNF dcreased;thephagocytosis activity of Mφ from abdominal cavity dcreased;and the activityof NK cell dcreased;the GLU,CHO,TG increased obviously;the activityof SOD dcreased obviously and the activity of MDA increased obviously.D-galactose induced aging mouse model have the similar changes of 16-24months old mouse,So it can be used in the research ofimmunosenescence .Otherwise,the main purpose of this paper is to research ifthe protection effect of VE exist and the machnism of VE on aging inducedby D-galactose. In the rats clysters by VE, results indict that : the ThymusIndex increased;the aptosise of thymus cells decreased;the proliferation of Tlymphocyte increased;and the activity of cytocine IL-2,INF-γ and TNFincreased;the phagocytosis activity of Mφ from abdominal cavityincreased;and the activity of NK cell increased .From these ,VE couldmediat the immune system function of aging model mouse.On the other hand,cmpared with model control, the activity of SOD increased obviously and theactivity of MDA dcreased obviously in aging rats were clysterd by VE. Fromthis tow index ,VE could protect aging model mouse to the damage of freeradical,and this function maybe the key of VE to pretest aging model mouse toimmunosenescence. These studies offer use VE therapeutic approaches towardimmunosenescence of clinic, and provide new experiment provement ofpretest immunosenescence.
