| In this research 5 compounds were isolated from the stems of Marsdenia tenacissima by modern technologies of extraction,isolation,purification,identification.The structures of 4 compounds have been identified on the basis of physico-chemical and spectrographic evidence.They were dihydrosarcostin ( â… ), α -amyrin acetate (â…¡), β -sitosterol (â…¢) , stearic acid (â…£).Compound â… was isolated from the plant for the first time. In the extraction process,the extraction conditions of Marsdenia tenacissima were determined by the orthogonal test and the content of total steroidal glycosides as index was detected by the UV spectrophotometry.The optimal conditions are as followed: 90% ethanol, extracting for 2 times ,ratio of solvent volumn and the stem weight is 10:1 and 8:l,and the extracting time is 1.5 and 1.0 hr respectively. With macroporous absorbing resin HPD100,the active parts of ethamol extracts were refined.We optimized the best absorption conditions by orthogonal experiment.The eluation solvent has been investigated.The result was that 70% ethanol was superior to 30%, 50%, 90% ethanol.By orthogonal experiment,the best desorption process has been established.Under the guidance of the theories of modern pharmaceutics, Tong-Guang-Teng capsule has been reseached .All of these indicated that the prepration procedure was scientific ,reasonable and practicable. |
