| Objective To observe the preventiont effect of berberine on the carotid artery atherosclerosis in rabbit.Methods Thirty-two Japan white rabbits were assigned into four groups randomly:Normal group,Control group,Simvastatin control group and Berberine prevention group.The rabbits in the normal group were fed with normal diet; the rabbits in the control group were fed with high cholesterol-diet for five weeks,in which the rabbits were operated by air-dry on the right common carotid artery after a week;the rabbits in the simvastatin control group were fed with high cholesterol-diet and took simvastatin orally for five weeks,in which the rabbits were operated by air-dry on the right common carotid artery after a week;the rabbits in the berberine prevention group were fed with high cholesterol-diet and took berberine orally for five weeks,in which the rabbits were operated by air-dry on the right common carotid artery after a week.After five weeks,all the rabbits were done the carotid artery DSA and the pathological examination and caculated the sum of macrophage cell by immunohistochemistry.Results The carotid artery in rabbits was found stenosis in the control group, but not observed stenosis obviously in the berberine prevention group and the simvastatin control group by DSA, The degree of carotid artery stenosis in the control group was (60.11±13.02)%, which was (28.22± 12.66)% and (29.45± 14.03)% in the simvastatin control group and the berberine prevention group respectively;Compared the degree of stenosis in the control group with the simvastatin control group and the berberine prevention group,there was signifficent difference between the control group and the berberine prevention group or the simvastatin control group (P<0.01),but not between the simvastatin control group and the berberine prevention group (P<0.05) .There was notable atherosclerotic plague in the control group, but found early atherosclerosis plague in the simvastatin control group or the berberine prevention group. The I/M of carotid artery in the control group was 1.11±0.007, that was 0.51±0.005 and 0.55 ±0.007 in the simvastatin control group and the berberine prevention group respectively,there was signifficent difference between the control group and the simvastatin control group or the berberine prevention group(P<0.01) ,but not between in the simvastatinc control group and the berberine prevention group (P>0.05). the sum of the macrophage cell was 66.12+3.88 in the control group,which was 35.20+3.45 in the simvastatin control group and 38.32±3.40 in the berberine prevention group.the sum of macrophage cell in the control group was more than that in the simvastatin control group or the berberine prevention group (P<0.01),but not significant different between the simvastatin control group and the berberine prevention group (P>0.05).Conclusion Berberine had the prevention effect on the formation of the carotid artery thaeosclerosis. |
