Objectives: To study the effects of ITF on TNF-a(tumor necrosis factor-α) and NO(nitric oxide )density in neonatal rat NEC model, to discuss whether if ITF has protective function in NEC; To investigate the intracellular distribution and the activation of ERK in neonatal rat intestinal tissues and the role in the mechanism of NEC.Methods: Forty neonatal rats are divides randomly into five groups,(l)NEC+ ITF 0.5mg, (2)NEC+ ITF 0.2mg, (3)NEC+ N.S0.5ml, (4)NEC+ N.S0.2ml,(5)all others. NEC model of neonatal rats is established as followed .One-day old Wistar rats are put into 100%carbon dioxide for 5 minutes and then 100% oxygen for 5 minutes. After that, they are returned to their mothers.This procedure is done once everyday and continued 3 days. On the 4th day ,all the subjects are put to death .Intestinal tissue locates at the boundary of ileum and cecum is obtained to observe histological changes and ERK level. Other intestinal tissue is turned to homogenate. After the homogenate iscentrifuged,supernates are used to test the density of TNF-a and NO.Results: The density of TNF-α (pg/mg·pro) are significantly decreased in group A and B than those in group Cand D,(P<0.01);which have no difference with those in group E, (P>0.05 ) ;The density of NO (μmol·mg-1 tissue)are significantly decreased in group A and Bthan those in group Cand D, (P<0.01);which have no difference with those in group E , (P>0.05 ) . The density of ERK are significantly increased in group A and B than those in group C and D, (P<0.01);which have increased in group C and D than those in group E, (P<0.05); Activation of ERK with evident nuclear translocation was found in hypoxia-induced in neonatal rat NEC model. And more, ERK, TNF-α and NO content between group A and B, group C and D is not dramatically different .The pathological lesions indicate that intestinal tissue necrosis is severe in group C and D,which was graded 3 point, but obviously lessen in group A and B,which was graded 1 point, with ITF interfering.Conclusions: Intestinal inflammation was ameliorated after ITF was injected hypodermically or intraperitoneally .ITF may provide a brand-new way for the therapy of NEC in neonatal rats; ERK signal pathways might play important roles in the mechanism and signal transduction of NEC; may ITF protect the intestinal injury of neonatal wistar rat NEC model by activation of ERK signal transduction pathsway.
