| Kunming mice erythrocytes were used to prepare carrier erythrocytes loaded with VCR using a modified hypotonic pre-swelling and isotonic resealing technique. The entrapment rate and recovery rate of mice blood VCR-loaded erythrocytes are good. The total VCR-loaded amount in erythrocytes was (62.99+2.48)%. and erythrocytes recovery rate was (76.61+5.37)%. VCR-loaded erythrocytes have obvious inhibitive effect on S180 sarcoma, the average tumor weight was(0.33+0.05)g and the inhibition rate was (42.69+4.35)%. It is superior to use free VCR under the same dose, in addition, it also can decrease the toxicity and side effects of the drug. Furthermore, the inhibitive effect in tumor vaccine plus Calmette-Guerin Vaccine was also visible, which the inhibition rate was (42.63+24.22)%. In vivo distribution of VCR-loaded erythrocytes proofs that the VCR-loaded erythrocytes is more advantageous and effective to tumor therapy than given VCR independently. Erythrocytes can be used as drug delivery system of antineoplastic agent and has wide prospect in the clinical application. |
