Objectives:The pathogenesy of nonalcoholic fatty liver diseases (NAFLD) is not yet identified, researchers consider it is related with metabolic syndrome, such as obesity, hypertention, diabetes and hyperlipoidemia. It is generally accepted that peroxisome proliferators activated receptorγ (PPARγ) plays an importand role in regulating the metabolism of lipid. To study the association of PPARγ2 gene polymorphism with NAFLD, we carry out this study. Our research object is the Han nationality population in Tianjin. There are 155 NAFLD patients and 141 control subjects in our study. The aim of this study is to investigate the prevalence of the PPARγ2 Pro 12Ala polymorphism by PCR-RFLP technique. In addition, our aim is to find out the correlation between the polymorphisms and NAFLD in the Han nationality population in Tianjin of China.Methods:1. To collect the blood samples of nonalcoholic fatty liver disease patients and control subjects.2. To extract and to purify the genomic DNA from the collected blood samples.3. Using PCR-RFLP technique, the PPARγ2 polymorphism site was screened in155 nonalcoholic fatty liver disease patients and 141 controls.4. To sequence the polymorphic gene found by RFLP technique.5. To detect and analyse the clinical data, such as FPG, blood-fat,FINS, et al. Results:For the PPARγ polymorphism site of Pro12Ala, 13 (8.39%) with the heterozygote mutation were found in 155 NAFLD patients. The A allele frequency is 4.19% in NAFLD. 11 (7.8%) with the heterozygote mutation were found in 141 controls. The A allele frequency is 3.9% in control group. There is no significiant differences between two groups in the A allele frequency (P >0.05). In both NAFLD group and control group, BMI, diastolic blood pressure , TG and the waist circumference in femail are significiant higher in PA genotype subjects than that in PP genotype subjects(P<0.05). In NAFLD group, we subsequently compared the clinical data between PA and PP genotype subjects, and found there is significiant differences in ALT between this two genotypes (P <0.05).Conclusions:In the Han nationality population in Tianjin of China, Prol2Ala polymorphism of PPARγ2 gene may not play a critical role in the development of NAFLD. But it participates the regulation of some metabolism objects, such as TG, BMI and diastolic blood pressure. In NAFLD group, it may be the reason of progression to liver inflammation.
