Bile duct carcinomas are associated with gloomy prognosis. In recently years, the incidence showed an ascending trend because of attention of people and improvement of imaging technologies. The most familiar diagnostic pattern was as follow: jaundice + dilation of bile duct + mass of bile duct, while the jaundice frequently indicated terminal stages of cancer. So whether or not early diagnose bile duct carcinomas became one of the most important factors affecting prognosis of this disease.Proteomics analysis technology is a new, high throughput post-genomic research method, which has been widely used in studies of oncology. One of these important studies is screening out some proteins which have specially changed in serum of bile duct carcinoma patients, and then judging if they could be used as diagnostic tumor markers.Through a retrospective analysis of diagnosis course of 60 bile duct carcinoma patients from 2004 to 2006, we investigated possible related factors and sought feasible methods for diagnose bile duct carcinoma early. Meanwhile we primarily compared serum proteins between bile duct carcinoma patients and benign bile duct carcinoma disease patients. PBSII-C protein chip reader and screened out some proteins that expressed differently. Finally the feasibility of them as diagnostic tumor markers was estimated.The results were as follow:1. 52 patients were diagnosed after jaundice had appeared while jaundiceusually indicated the course of diseases had been into end stage.2. Up to the present there is no serological parameter with high sensitivity and specificity for diagnosing colorectal cancer.3. Compared with the spectra of benign bile disease patients' serum there were 10 potential markers detected in the spectra of the bile duct carcinoma patients' serum, among which the proteins expression were high in 3 and low in 7.Conclusion:1. It is very difficult to diagnose bile duct carcinoma early.2. Up to the present there is no serological parameter with high sensitivity and specificity for diagnosing colorectal cancer.3. There are lots of proteins expressed abnormally in bile duct carcinoma patients' serum. Some of them are potential for diagnostic tumor marker in detecting bile duct carcinoma.
