The contrast research was done to observe the effect of polysaccharide sulfate (PSS) amlodipine(Aml) and benidipine (Ben) on renal hemodynamics in dogs. The effect of PSS and Ami on urine flow, urinary excretion of sodium and potassium and glomerular filtration rate were measured in rats, all of which increased urinary excretion. PSS and Ami were administered arterially into denervated kidney in dogs, followed by measuring several renal hemodynamic parameters, including glomerular filtration rate, urine flow, urinary excretion rate of sodium and potassium and etc.The results showed that both PSS and Aml significantly increased renal blood flow and urine flow(P<0.01).As for other parameters, PSS was similar to Aml, except for weaker effecacy and shorter persistent period . suggesting that the effect of PSS was relatively mild.The effect of PSS and Aml on NO and cGMP in rat renal tissue were also measured. The result showed that, instead of PSS, Aml could increase the concentrations of both NO and cGMP, while dilating renal arteries to some degree. It was suggested that PSS increase urinary excretion possibly by inhibiting the renal tubular reabsorption, while Aml increase glomerular filtration rate to realize this. By observing the effects of PSS, Aml and Ben on renal afferent and efferent arterioles with microdialysis technique, we knew that Aml dilated afferent arterioles while constricted efferent ones, Ben dilated both whereas PSS showed no effect, which may provide useful information when administering Ca antagonists and PSS to decrease untoward reaction.
