| Objective The expressive level of connexin 43 (Cx43) after Bladder Outlet Obstruction (BOO ) on detrusor was studied in order to elucidate the pathogenesis of detrusor instability (DI).Methods Animal models of BOO were made in wistar rats, which were divided into five groups by the chronological order and the means of cystometry. RT-PCR and Western blot technique were employed to detect the expressive level of Cx43. Results The rate of DI occurrence was 65% after BOO 6 weeks. There were no Cx43 mRNA expression in the control group and no significant difference in other groups (p>0.05). The level of connexin 43 mRNA expressions in DI groups was higher than that in other groups (p<0.01). Positive rate of Cx43 protein in DI group was significantly higher than that in other groups (p<0.01).Conclusion It is a kind of protective reaction that the expression of the Cx43 after BOO , but such high expression in DI group is perhaps decompensation. It is suggested that we can find a method to cure the DI by interfering in the high expression of Cx43. |
