Objective:This paper deals mainly with studies on chemical constituents and activity of corydalis saxicola Bunting. To explore the anticancer activity of total alkaloids of Corydalis saxicola Bunting is the major work. The proliferation inhibitory effect of total alkaloids of Corydalis saxicola Bunting was investigated in five tumor cell Lines.Method : Various column chromatography and preparative TLC were employed for the isolation and purification of constituents. Chemical structures of the compounds obtained were elucidated by the spectroscopic methods.Five cancer cell lines: CNE-1, CNE-2, A2780, SKOV3 and PM2 cultured in vitro. Proliferation inhibitions of five cancer cell lines were tested by MTT assay. The morphologic changes were studied by HE dyeing. Results Ten compounds were isolated by silica gel column chromatography, Vacuum Liquid Chromatography Method. Cholesterol, daucosterol, Berberine and coptisine were isolated and identified from Corydalis saxicola. MTT assay showed that total alkaloids of Corydalis saxicola might significantly inhibited proliferation of five cancer cell lines, in concentration-dependent manner under the 15.63,31.25,62.5,125 and 250μg/ml.Morphologic observation presented that SKOV3 cell differentiation was induced by total alkaloids of Corydalis saxicola.Conclusions : The total alkaloids of Corydalis saxicola Bunting has cytotoxicity against the five cancer cell lines.The active effect of Corydalis saxicola Bunting was also studied.
