The Effects Of Pulsatillae Decoction And Its Single Components (Chinese Pulsatilla Root, Golden Thread, Amur Cork-tree, Ash Barkon) Liquid At Different Doses On Contractility Of Isolated Rat Ileum

OBJECTIVE: TO observe the effects of Pulsatillae Decoction(PD) and its single constituents:Chinese Pulsatilla Root (CPR), Golden Thread (GT), Amur Cork-tree (ACt), and Ash Bark (AB) liquid at different concentrations on contractility of isolated rat ileum smooth muscles and explore their probable mechanism.METHOD: The routine methods for experiment on contraction-effect of the isolated rat ileum were emplayed. The effects of the drugs against spontaneous contractions, contractions induced by submaximal concentractions of acetylcholine (Ach) and histamine (Hist), and concentrations induced by CaCl2 on rats'isolated ileum were observed with MS2000 computer system. RUSULTS: 1. PD and its single constituents: CPR, GT, ACt, AB liquid, and Ver inhibited the spontaneous contractions of Wistar rats'isolated ileum smooth muscles. IC50 determinations (mg·ml-1) except for Ver (μmol·l-1) were 8.36±0.36, 9.02±0.64, 3.5±0.34, 5.9±0.34, 8.96±0.21 and Ver0.27±0.13 respectively. 2. PD, CPR, GT, ACt, AB liquid, and Ver inhibited the contractions induced by submaximal concentrations of acetylcholine (Ach) and histamine (Hist). IC50 determinations (mg·ml-1) except for Ver (μmol·l-1) were 6.86±1.16and 5.97±1.34; 7.41±0.89and 8.69±0.71; 5.55±0.16and 2.25±0.18; 6.12±0.57and 5.14±0.37; 8.52±0.31and 7.27±0.21; 0.58±0.15 and 0.4±0.22, respectively.GT had the strongest inhibition action than any other herbs. Act was stronger than the others, and AB showed less sensitive than PD, CPR. 3. PD, CPR, GT, ACt, AB liquid, and Ver inhibited the contractions induced by CaCl2 with different concentrations which showed noncompetive antagonists. The extent of inhibition of 5mg·ml-1, 2.5mg·ml-1, 1.25mg·ml-1GT liquid on Cmax of CaCl2 was 28.9%±5.26, 54.36±1.13,67.48±5.36; the extent of inhibition of 20mg·ml-1, 10mg·ml-1, 5mg·ml-1Act was 28.3%±2.0, 50.85%±3.42, 73.7 3%±3.24; the extent of inhibition of 18.33mg·ml-1, 9.17mg·ml-1, 4.59mg·ml-1PD was 38.28%±0.78, 55.16%±0.11,67.48%±5.36;the extent of inhibition of 16.8mg·ml-1, 8.4mg·ml-1, 4.2mg·ml-1CPR was35.11%±2.67, 43.15±1.13, 57.6%±0.61; the extent of inhibition of 20mg·ml-1, 10mg·ml-1, 5mg·ml-1 AB was 38.24%±1.78, 53.16%±1.33, 65.99%±5.36;and the extent of inhibition of 0.1μmol·l-1,1μmol·l-1,10μmol·l-1 Ver was 86%±5.21, 61.05%±1.1, 44%±5.36, respectively. 4. PD1.19mg·ml-1, CPR8.3mg·ml-1, GT0.75mg·ml-1, Act1.25mg·ml-1, AB10mg·ml-1and Ver1μmol·l-1 inhibited the contractions induced by Ach in Ca2+-free medium, while PD36.6mg·ml-1, GT15mg·ml-1, Act40mg·ml-1but not AB40mg·ml-1, CPR40mg·ml-1,...
