Objectives: To investigate both effect and pathogenesis of intestinal endotoxemia on the formation of portal hypertension (PUT) of hepatic cirrhosis, and fuether to reveal the forming mechanism of portal hypertension.Methods: The hepatic cirrhosis model of rats is treated with complex factors (CCl4, alcohol, et al). The animal of male wistar rats are executed at the end of 0,2,4,6,8 weeks. The PVP is observed by puncturing cranial mesenteric vein. The dynamic levels of plasma endotoxin, ALT (alanine aminotransferase), ET-1 (endothelin-1) and intestinal homogenate NO (nitrogen monoxidum) respectively are showed the changes. The stages of hepatic fibrisis are defined by staining HE (haematoxylin-eosin) and VG (van gieson). The dynamic expressions of TGF- β1 (transforming growth factor), α-SMA (a-smooth muscle agitated protein) and LN (laminin) in liver are observed by the method of immunohistochemistryResult: 1 The modeling of the hepatic cirrhosis of rats:By both the liver HE and V.G, it is confirmed that 2 weekend is the stage of fatty liver, which has a serious degeneration and necrosis comparing with other stages, and 4 weekend is the stage of hepatic fibrosis, and 6 weekend is the initial stage of hepatic cirrhosis, and 8 weekend is the stage of hepatic cirrhosis.2 The formation of IETM (intestinal endotoxemia):The result of plasma endotoxin shows a tendency to increase at 2, 4, 6 weekend compared with the control, and at 6, 8 weekend maintains a high level. There is a statistics difference existing between groups of model and control, P<0.05.3 The result of PVP:The result of PVP shows a tendency to increase gradually at 2, 4 weekend compared with thecontrol, but to decline at 6 weekend, then to increase gradually at 8 weekend. It is possible thatthe development of bypass circuit makes PVP decline at 6 weekend. There is a statisticsdifference existing between groups of model and control, P<0.05.4 The results of reflecting resisting force of hepatic sinusoid:The immunohistochemical results of a-SMA and LN confirm that the number of activated HSCand deposited LN at hepatic sinusoids show a tendency to increase at various time points, and...
