Objective:To establish a complex rat model of Alzheimer's disease, to investigate the functional mechanism and remedy effect of JiannaoYizhiTang on learning and memory,cholinegic system, oxidative stress and apoptosis of the AD model rats.Methods:1,Establishment of AD rat model. Injecting accumulation Aβ25-35 into the left lateral ventricle and the old Wistar rat to make complex AD rat model.2,Research of the functional mechanism and remedy effect of JiannaoYizhiTang on AD rat model. To Select male normal old rats, that was randomly divided into normal group,model group,Acript group and big,moderate,small dose of JiannaoYizhiTang group. Each group was lavaged with regulated dose ,After a week,except the normal group, other groups was injected 5μl accumulation Aβ25-35 in the left lateral ventricle. Seven days later ,we tested learning and memory ability of the rats by Morris water maze .After 3 weeks, to take the blood from abdomen aorta, then measured AchE,BchE and SOD activity in the serum, and then to get the brain, and selecte the left frontal lobe,the temporal lobe and hippocampus for toimmunohistochemistry and Bielschowsky dyeing,separating the right side pro-1/4 cerebral cortex,detecting apoptosis with flow cytometry;taking right side post-1/4 cerebral cortex to measured content of MDA,NO and activity of SOD.Results:1,The model group showed that the escape latency increased significantly compared with normal group ,Furthermore, the result of immunohistochemistry revealed that Aβdeposited inside of glia cell, similar SP of brain tissue, and CAA of cortex and result of silver staining showed NFTs and a number of glia cells.2,The Morris water maze test manifested: big, moderate dose of.JiannaoYizhiTang group and Acript group have shorter the escape latency than the model group( P<0.01), moreover, there is no significant difference between 3 goups, and the percentage of terrace quadrant swimming path length occupying total path length is the longer than that of the modle group,which has the obvious difference( P<0.01).3,The activity of AchE,BchE in the modle group is obviously higher than that in normal group ( P<0.01),and Acript group has the most significant function of decreasing activity of AchE, while each using medicine groups were no difference in decreasing BchE activity.4,The model group has the typical free radicals changes, the MDA content increased ( P<0.01),... |
