The Activities Of Anti-osteoporosis And Effects On Uterus And Mechanism Of A Novel Estrone-rhein Hybrid Compound | Posted on:2007-06-15 | Degree:Master | Type:Thesis | Country:China | Candidate:X Jin | Full Text:PDF | GTID:2144360185452985 | Subject:Pharmacology | Abstract/Summary: | PDF Full Text Request | Objectives: To study the effect of a novel estrone-rhein hybrid compound -estrone linked to rhein with piperazine as the bridge(Lc-1) on the bone mass, strength, texture, the proliferation of uterus, the expression of estrogens receptor subtypes(ERαand ERβ) and c-fos, c-jun proteins in endometrium in OVX rats, the effects of Lc-1 on the proliferation and differentiation of primary cultured osteoblasts of newborn rats for evaluating the activities of anti-osteoporosis and effects on uterus and investgating the relative mechanisms. Methods: (1)Sixty 3-month-old female Wistar rats were randomly divided into five groups:①Sham group②OVX group③H-Lc group (OVX + Lc-1 2.568mg/kg·d)④L-Lc group (OVX + Lc-1 0.514mg/kg·d)⑤E group (OVX + estrone 1.07mg/kg·d). Rats in the sham group were subject to sham operation. Ovariectomies were performed in the other groups. Administration of drugs oral one week after operation was carried out in the later six months. The sham and E group received vehicle only. (2)The body mineral density was determined before sacrifice. Then all rats were put to death. Serum samples were collected to detect Ca, P and ALP...
