We have linked the chemical construction of rhein, the ingredient of rhubarb anthraquinone,with that of traditional antineoplastic agent Podophyllotoxin , to obtain rhein derivatives RH-01. During the process of preliminary screening, we have found that RH-01 has a most powerful inhibition to the growth of human osteosarcoma (HOS) cell line, and showed a typical dose~dependent relationship in the assay.The aim of the exprements is to investigate RH-01's inhibition to HOS cell in vitro and to transplantable tumor in mice, and to discuss basically its mechanism of action.Method: HOS cell was seeded in a 96~well microtiter plate for 24h, then was treated with different concentrations of RH-01(0.05~0.8μmol/L), after cultivating for 96 hours at 37℃. The effects on cell growth were examined by MTT assay in vitro. Through MTT assay, we also measured the value of optical density (OD) of RH-01 at 0h ,24h, 48h and 72h with different concentrations(0.05~0.8μmol/L).
