Abstract: Objective To investigate the expression of c-fos , PCNA(Proliferating Cell Nuclear Antigen), Bax and Bcl-2 on the myocardial ischemia/reperfusion(I/R) in rabbit and explore their significance. Methods The model of local myocardial I/R in rabbit was established (aim to simulate myocardial ischemia/reperfusion area at the earlier period of operation in off-pump coronary artery bypass grafting surgery) , eighteen rabbits were randomly divided into the three groups (n=6), ischemia 15 minutes followed by 0 min reperfusion group (group I); ischemia 15 minutes followed by 15 min reperfusion group (group II); ischemia 15 minutes followed by 30 min reperfusion group (group III). specimen taking from left ventricle myocardial I/R area and control area( right atrium & ventricle, without I/R process ), observe the change of HE staining, the expression of c-fos ,PCNA,Bax and Bcl-2 by using immunohistochemistry method, the result were analyzed by SPSS 11.0 software. Results the expression of c-fos gene , PCNA , Bax protein and Bcl-2 protein on the myocardial ischemia/reperfusion were founded in all of groups, ischemia/reperfusion can increased the percentage of the expression of c-fos gene, PCNA, Bax protein and Bcl-2 protein compared with each other and those in control group. the increase of the expression of c-fos gene enhances the ratio of Bax/Bcl-2. Conclusion (1) Ischemia and (or) reperfusion can stimulate the expression of c-fos gene, after reperfusion the percentage of the expression increased; (2) The increase of c-fos gene expression correlat with myocardial ischemia/reperfusion injury(MIRI); (3) After myocardial ischemia/reperfusion, the expression of c-fos gene probably related to regulating balances of cell death/survival process, c-fos gene maybe startup the expression of apoptosis gene , anti-apoptosis gene and proliferation gene, the c-fos gene maybe startup the earlier period apoptosis and protective reaction after MIRI. (4) c-fos gene detection may become an important method for diagnosis of MIRI.
