It was thought that dental pulp stem cells (DPSCs) were progenitors existing in dental pulp. They could be activated by dental damage and terminally differentiated into functional odontoblasts. Insulin-like growth factor-1(IGF-1) and basic fibroblast growth factor (bFGF) were important poly-peptides and mitogen, which promoted proliferation and differentiation in the growth of cells and played an important role in the development of teeth. Studies showed that IGF-1 had been implicated in the proliferation and differentiation of dental pulp cells; bFGF could significantly stimulate proliferation of dental pulp cells cultured in vitro, so they have been paid more and more attention. It has been suggested that aFGF or bFGF is not be able to induce differentiation of dental papilla into odontoblast alone. bFGF could act synergistically with IGF-1. Been combinated of IGF-1 and bFGF could promot the surface cells of dental papilla polarization and functional differentiation into odontoblast, as well as extracellular matrix deposition formation. Combination of IGF-1 with aFGF caused pronounced cell elongations but a little matrix deposition; Combination of aFGF or bFGF with TGF-β1 caused cell polarization at the surface of dental papillae, but matrix secretion was restricted to a few explants. Simultaneous addition of IGF-1 and bFGF could strengthen their inductive effects and promote functional differentiation of dental papillae into odontoblast cells.Whether IGF-1 or bFGF could enhance the proliferation and differentiation of human dental pulp stem cells had been poorly investigated. In this study, we preliminarily cultured dental pulp stem cells and explored the effects of IGF-1 and bFGF on the proliferation and differentiation of dental pulp stem cells. It provided experimental evidence for further research of proliferation and differentiation of human dental pulp stem cells.
