Objectives: This prospective study aimed to evaluate the efficacy and drug resistance of continuous infusion (CI) versus intermittent administration (IA) of ceftazidime in a Klebsiella pneumoniae rabbit pneumonia model.Methods: Forty Klebsiella pneumoniae pneumonia rabbits were randomized to one of five groups (n = 8 for each group). Group A and B received 10mg/kg and 20mg/kg of ceftazidime by intravenously CI. Group C and D received 5mg/kg and 10mg/kg of ceftazidime every 12 hours by intravenously IA. Group E was not infused as control. For the five groups, BAL samples and venous blood samples were collected in different time points from the start of administration for MIC and SIT/SBT test. The chest X-ray examinations were made after inoculated 24h and at the end of treatment. Then the animals were killed for lung pathologic examination. The plasma samples were performed drug assays by HPLC and SIT/SBT by microdilution method. In vitro susceptibility test was determined by the microdilution technique.Results: The CI of 10mg/kg/24h and 20mg/kg/24h of ceftazidime resulted in a mean steady-state concentration in serum 1.308±1.409ug/mL and 2.691±1.384ug/mL, respectively. The IA of 5mg/kg/12h and 10mg/kg/12h of ceftazidime resulted in a mean peak concentration in serum 10.51±3.13ug/mL and 14.61±9.61ug/mL, and a mean trough concentration in serum 0.06±0.09ug/mL and 0.33±0.26ug/mL, respectively.
